Grape seed extract is a kind of polyphenol extracted from grape seeds, which is mainly composed of proanthocyanidins, catechins, epicatechins, gallic acid, etc.
1. Antioxidant capacity
Grape seed extract is a pure natural substance, and is one of the most efficient antioxidants from plant sources found so far. The test shows that its antioxidant effect is 30-50 times that of vitamin C and vitamin E.
2. Activity
Proanthocyanidins have extremely strong activity and can inhibit carcinogens in cigarettes. The ability to capture free radicals in the water phase is 2 to 7 times that of general antioxidants, such as more than twice the activity of α-tocopherol.
1. Grape seed extract can be used in the field of medical care, protecting the heart and blood vessels and preventing diseases.
2. Grape seed extract is one of the most effective antioxidants, which can maintain skin elasticity and moisture, anti-aging and anti wrinkle. Therefore, it is widely used in the field of cosmetics.
3. As a natural preservative, grape seed extract is widely added to cakes, cheese and other foods to improve food safety. At the same time, grape seed extract can be made into capsules, tablets, granules and other health food.
The study found that the content of proanthocyanidins in grape seeds and pine bark extracts was the highest among many plant tissues, and the main methods of extracting proanthocyanidins from grape seeds were solvent extraction, microwave extraction, ultrasonic extraction and supercritical CO2 extraction. The grape seed proanthocyanidin extract contains many impurities, which need further purification to improve the purity of proanthocyanidin. The commonly used purification methods include solvent extraction, membrane filtration and chromatography.
The ethanol concentration had the most significant effect on the extraction rate of grape seed proanthocyanidins, while the extraction time and temperature had no significant effect on the extraction rate of grape seed proanthocyanidins. The optimum extraction process parameters are: ethanol concentration 70%, extraction time 120 min, material to liquid ratio 1:20.
The static adsorption experiment showed that the adsorption rate of HPD-700 to proanthocyanidins was 82.85%, followed by DA201, which was 82.68%. The difference was not significant, and the adsorption capacity of these two resins to proanthocyanidins was also the same. In the desorption test, the desorption rate of DA201 resin to proanthocyanidins was the highest, 60.58%, while the desorption rate of HPD-700 to proanthocyanidins was only 50.83%. Combining adsorption and desorption experiments, DA210 resin was determined as the best adsorption resin for the separation of procyanidins.
Through process optimization, when the concentration of proanthocyanidins is 0.15 mg/mL, the flow rate is 1 mL/min, 70% ethanol solution is used as eluant, the flow rate is 1 mL/min, and the amount of eluant is 5 BV, the extract of grape seed proanthocyanidins can be preliminarily purified.
